Dynal Biotech has announced two product releases from its new
proteomic preparation range, which use magnetic bead chromatography
technology to prepare for mass spectrum analysis without the
requirement of columns.

The weak cation exchange (WCX) Dynabeads and reversed phase chromatography (RPC) Dynabeads systems have been designed to be a viable alternative to chromatography columns, spin columns and pipette sample preparation technologies. They can fractionate mixed protein samples for downstream analysis such as mass spectrometry.

Dynabeads WCX and Dynabeads RPC Protein can be used to concentrate, prepare and desalt protein mixtures. They are specifically designed for the fractionation of intricate mixtures of protein (e.g. serum, plasma, urine or cell lysates) based on different parameters: Dynabeads WCX carry surface charges, which reversibly adsorb oppositely, charged proteins and large peptides. Dynabeads RPC Protein has a coating with optimised hydrophobic and hydrophilic components for peptide/protein binding.

Magnetic Bead Chromatography is unique in that it exhibits magnetic properties only when placed within a magnetic field. When added to a heterogeneous suspension, the Dynabeads will bind to the desired target (cells, nucleic acids, proteins or other biomolecules).

This interaction is based on the specific affinity of the ligand on the surface of the Dynabeads. The resulting target-bead complex can be removed from the suspension using a magnet (Dynal MPC). Bead-bound complexes are drawn to the side of the tube facing the magnet, and the supernatant is removed with a pipette.

The advantages of magnetic handling allow for easy washing, separation and concentration of the target without any need for centrifugation or columns. The lack of magnetic remanence combined with the excellent dispersion abilities also meet the demands required for automated systems and microfluidics.

Fractions are then released via controlled step-wise elutions. For Dynabeads WCX this is achieved using increasing concentrations of salt or by changing the pH. Dynabeads RPC Protein use hydrophobic interactions and therefore increasing the concentration of acetonitrile will release protein fractions.

Since the beads use different properties the two can be used to isolate different fractions from the same sample. This means that they provide an adaptable preparation protocol for downstream mass spectrum analysis, HPLC and 1D/2D gel electrophoresis.

Dynal Biotech Dynabeads remain dispersed in solution but move in response to a magnetic field, with no magnetic remanence. The beads have been designed to accommodate different surface chemistries and have been developed in a number of different sizes to suit different genomic, proteomic and in vitro diagnostic applications.
